Principle
As a kind of immunoassay, FIA also has two modes, namely competition type and sandwich type. Among them, the competitive type (take the labeled antigen competitive type as an example) is based on the immunoassay method based on the competition between the unlabeled antigen (Ag) and the labeled antigen (Ag-L) to bind to the limited antibody (Ab). For sandwich immunoassays, the reaction principle is to immobilize excess Ab on the immune response carrier, and then add a certain amount of Ag. After the immune response, add excess labeled antibody (Ab-L) to form a "sandwich" "Sandwich immune complex. The more Ag present in the sample and the more Ab-L bound, the stronger the labeled fluorescent signal of the sandwich immune complex.
